Recently I ended up trying Gnome 3, and I've stuck with it. I like the no-fuss way it looks (one panel, with not too much on it) and it seems quite intuitive to me.
As mentioned by notthatguy and bmc5311, it is well worth reading the cheat sheet to get up to speed.

I also like the way it is equally well set up for mouse-only or keyboard-only use.
I prefer the keyboard usually: use the "Super/Windows" key to get to the Overview, then just type the first letter of the program you want and hit <Enter>.
"i" for internet (Iceweasel)
"f" for files (Nautilus)
"t" for terminal
"v" for video (VLC)

..and if the program you want is not the first one that comes up when you type the first letter, just choose it by using the down arrow. Then next time you type that letter, it will appear first on the list :D

And that's before you even start to customise it... suits me fine so far.

Changed vote to KDE, because that's what I'm using now.

Openbox on my old desktop. Very plain, no icons. Gnome 3 on my laptop. It's new and can handle the resource requirements. I wanted to try it and will keep it for another month or two. Still, of the two, I prefer the Openbox. Eye candy wears off quickly.

First I switched to KDE4, used it for about a month then switched to XFCE since the start of the year.

In XFCE I have something simple and clean that works and doesn't get in my way. There are things that bug me about Thunar and Xfwm, but I can ignore them. I already learned to ignore and figure out workarounds to a bunch of stuff using KDE4.
